This simple high-throughput system measures individual respiration in tiny organisms like zebrafish embryos, Daphnia, fish/invert eggs or larvae, Drosophila etc., based on a 24-channel optical fluorescence oxygen reading device. Individual organisms are placed inside gas-tight 80-1700 µl glass chambers sealed with an oxygen impermeable PCR film for real-time oxygen consumption rate measurements. The glass material provides non-permeable surfaces, unlike ordinary well plates that are made of gas permeable plastics. Each chamber is fitted with a non-invasive and re-usable oxygen sensor spot for dynamic fluorescence quenching oxygen sensing. The sensors can be re-calibrated and sterilized for multiple trials. The entire setup can be placed inside an incubator for temperature control, or use our optional flow-through water bath and a circulating bath. A transparent self-adhesive sealing film is applied on top of the microplate to seal all 24 chambers in one go.
